Another solid resource would be the ComicsPro Message board. There is an $80 price to join, but you get mentoring from established retailers. It's designed for people that want to open a comic book store and need a whole bunch of advice.

At one time, I wanted to own my own comic book store. Today, I'd be very hesitant to open a new store. In my area, Toledo, Ohio, there are 3 existing comic book stores. If I was going to get into the business, I would be quite willing to buy any of the 3. All 3 of them have been there for around 20 years, and I see ways to improve all of them.

In the big picture, the single most important thing for opening a new store or buying an existing store is to have money, lots and lots of money. At a new store, you will be losing money for a long, long time. At an existing store, you will have to make big monthly loan payments.
